I think the reason why L said to assume Light is Kira is because only Light knows who L is. Other than the detectives he works with, no one else knows L. You need to remember L has essentially 3 names, L, Ryuga (Fake name) and His real name which we don’t even know. L already knows that the 1st Kira (who he suspects to be light) needs a name to kill but the 2nd one doesn’t. And for the 2nd Kira to kill L, the 1st Kira would need to tell them who L is. Outside L’s trusted circle only Light knows that he is L, he even said it when he was sitting on the bench.

L knew Misa was associated with Light because L had people trailing Light. We even saw the scene where they got spotted together, so once they found hair and evidence they were like okay so Misa is probably the second Kira and who was Misa just spotted with

To answer your question Charlie, L said im his explanation that chances are kira and the second kira are working together so he is fucked big time reason why he said the thing about if he gets killed in the days to come (which is expectable since to 2 kiras are working against him) then light is kira (not surprising either as he was his prime suspect all along anyway and everyone in the task force knows)

L is sure of Misa as second Kira right? then he saw Misa and Light together.... of course his next assumption would be, Light is Kira. - which he has always suspected before. and suddenly these 2 know each other? of course that would somehow give validation to his guess

“At this point in the story [when L tells Mr. Yagami that if he dies within the next few days, they should assume Light is Kira] did anyone know that Light had been meeting Misa?” Yes. In fact, the scene where L drops that bombshell on Mr. Yagami is partly a consequence of exactly that. A few days earlier, L had already told Mr. Yagami that he was going to have someone follow Light. The reason was Aoyama. Light had been in Aoyama on the 22nd, and then, on the 23rd, the Second Kira sent a tape saying, “I found Kira.” That was extremely suspicious. So Mogi starts following Light, both on campus and around town. He sees Light with Takada and assumes she’s Light’s “girlfriend.” Then he also witnesses Misa literally jumping into Light’s arms in the middle of the street and thinks, “Another girlfriend?” Mogi definitely found that suspicious. And this is where the series doesn’t actually show us every step of what happened afterward, probably because it wanted to preserve the surprise for the audience. But it’s pretty easy to piece together. L had already asked Aizawa to take the tapes Mr. Yagami recovered in for forensic analysis. They found hair, traces of food, dust, and fingerprints on them. The fingerprints weren’t Misa’s, as we already know, but the hair, food residue, and dust were connected to her. So when Mogi reported this “second girlfriend” suddenly meeting Light in the street, L must have had her apartment investigated as well. The forensic team would then have found hair, dust, and food residue there that matched what they had recovered from the tapes. L even tells Light about this at the university when he informs him that Misa has been arrested. So yes: Mogi knew Light had been meeting Misa, and by this point L was already actively investigating her. There’s another little detail that makes this even clearer. When Misa meets L at the university and L tells her that he’s “been a huge fan ever since her debut in Eighteen magazine”… come on. That makes absolutely no sense. Do you really think L sits around reading teen magazines? He says that because he had obviously INVESTIGATED Misa, not because he was actually some huge fan. It was basically a cover for that weird little look of delight he made the moment he saw her. And somehow even Light didn’t pick up on how strange that was. Maybe the series could have done a better job of showing every step of this investigation, but once you’ve watched it a few… actually, I’m not going to say how many times I’ve watched Death Note, because someone might put me in a straitjacket.

I think L found some hairs on the last day of the second. Kira sent. From her message he already suspected that the two Kiras have been in contact, but they were still waiting for the DNA results. If you notice, right after Light’s phone call, L gets a call, and that’s when they find out who DNA belongs to. So since he had literally met Misa just a few minutes earlier, that basically confirms to L that Light is Kira 👀👀

L set things up so that Light would be the prime suspect if he died because he is assuming the 1st and 2nd Kira were working together. Since Light is the only person L revealed that he was L to (outside of the task force) he wants to make it clear that if the second Kira somehow figures out that Ryuzaki is L, the only way the 2nd Kira could have possibly figured that out is if Light told them. The second Kira having that information at all makes Light extremely suspicious.
