No, Luffy couldnt conquerors haki anyone of the level of the tougher guys. He could make all the nameless characters go sleepy though. But coc haki can only knock out people significantly weaker than you. Or significantly weaker willpower? For example Rayleigh (well he probably wasnt giving it his best maybe? I dunno) couldnt knock out either of Laws closest crew members. And that's Rayleigh. So nah, Luffy couldnt do that. Its for clearing out scrubs, doesnt do anything against people that could actually tango a bit with you. While it would be handy, haki isnt a cheatcode you can use to take out opponents stronger than you as swell as that would be!

I always believed Raleigh knocked out specific people in that scene, because of the level he's at he could target individual people in that room instead of the sweeping blasts we normally see to take out fodder.
