No, Luke is right. The air would not rush out of the room just because it's not affected by gravity. Convection current would stop but that's about it

I mean, surely sending one disk is better than none?? Jolyne has no idea where the other disk is, or how long it'll take for her to find it. And it's been already established that there's only so much time left till Jotaro will start to decay That, and as we saw with Lang Ranger, the longer she carries the Stand disk around the bigger the risk that one of Pucci's goons will snatch it from her. And then she'd be right back at Step 1 (except it'd be unlikely she ever gets her hands on it again). Better not to take the risk

for the "pee" scene i think its to demonstrate that you never choose your timing for fight / hard situation , did you ever ask yourself when youre on a car or street and you're having to go to the toilet thinking "oh god if something happens i'm cook because i'm about to piss myself if i stop to help / talk right now " . i like to thi,nk that's what araki describe with this scene , bad timing happens
