Riyo isn't romantically interested in Rudo. When Riyo said, "Should have just let her be," it was meant to show that she didn't understand why, after everything the group went through because of Amo, Rudo was willing to apologize for his own behavior and even try to form a kind of friendship with Amo. When the group fought Amo, Riyo was one of those affected by her powers, and after Amo was defeated, Zanka reflected on how terrible Amo's ability actually was and how incredibly painful it could be for certain people and looked directly at Riyo, who looked defeated and sad. So, Riyo has clearly experienced something truly painful in her past. And as a manga reader, I can say that Riyo's past is still a mystery. The author, Urana, is really good at building mysteries around the characters. In my opinion, Riyo somehow sees herself in Rudo (for whatever reason) and therefore questions his decisions in particular. That's why, when she thought about a moment with her and Rudo, she wondered what would become of him. I'd say Rudo is like a little brother to Riyo, someone she's protective of. We saw in this episode, in particular, that the Cleaners are basically like Riyo's family, and she wants to protect them at all costs, even if it means getting her hands dirty.

If I return with an edit on this comment, then I fully caught up with them on their one piece, Mashle, this all their shows. I like when Ron reacts to the community, but that’s only a few have recent comments. I work a blue-collar job and I literally have no free time anymore. There’s just so much content I cannot keep up. I’m not complaining. I always have something I can watch now but man I’m trying to binge this to get caught up with them just to do it all over again next week.

i think when intially thinking of rudo, she still doesnt know how rudo is going to end up. Like if he will be harm to riyo's team or not. So initally being around him so much was to see if she needed to kill him or not. Not fully sure about the amo one, could be jealousy or her finally looking out for him.
