Koro-sensei is not just teaching the kids how to improve their education and assassination skills...

When Irina's old mentor shows up to tell her that she is no longer capable of being an assassin, she and Lovro are now given an challenge to see who can "kill" Karasuma first.

Koro-sensei continues on later saying that Irina has taken on the job of being a teacher, but she is teaching the kids how to kill with different skill sets. He praises her growth!

I might be wrong, but I think you are incorrect on one point, Koro-sensei isn't supposed to take over the world, but destroying it like he destroyed the moon, that why even if they like him, it's still a mission to take him out before he destroy our planet. I don't remember exactly what kind of info we will have with the brother, but you might start to have some answers and you are right to be more interested at how Koro become like this, that is part of why the show is good, but to be good enough to last two season, you need to sprinkle that kind of infos, so be patient and bear with it ^^.
