Actually Grimmjow didnt tell her to heal the 2 girls, he didnt care about them, Orihime just did it. So, it wasnt the first time.

Orihime did heal enemies before if im correct, I believe Orihime healed Loly and Menoly without Grimmjow's orders. In fact, Grimmjow explicitly commanded Orihime to heal her own facial bruises first, but she chose to ignore his request and instead rushed to save her two tormentors. edit: She also protected maki maki in souls society arc.
