1:25 as the episode two suggested this GIlgamesh is after his quest of finding the immortality herbs, which according to the epic poem he has learned something after his success, something that his young self always ignored and find it foolish(There will be more about this in the shows so i'll wait you see it till the end). We can basically say he has some development diverge from what he's become in FSN he still a bitches sometime but a lot better, because if you notice Gilgamesh in Fate Zero despite being a jerk he still respected Iskandar in his own way but Gilgamesh in FSN? totes piece of shit, probably due to the end of Fate Zero after having a contact with the content of holy grail(the black mud) making him disappointed and despise human.

23:20 Enkidu in epic of Gilgamesh was supposed to be man created from clay, to be a companion and equal to Gilgamesh despite his appearance is somewhat looks animal-like and wild. In the poem even mentioned Enkidu has sexual intercourse with Shamhat, a sacred temple prostitute, for six days and seven nights some translation say two weeks before Enkidu learn of human society and transforms into a civilized human, but either way, in the legend indicate that Enkidu was a male. while in Fate, his origin didn't changed much he's still made of clay and and makes him genderless, but he can modify his body and transform at will, so he can become both man and woman if want to. Another thing is his appearance, in Fate it's more monster-like than animal when he's first create, he still met Shamhat but sexual intercourse didn't mentioned but he has transform himself into human by took the appearance of Shamhat, that is the reason why his appearance in Fate looks like female. Gilgamesh use men pronouns for Enkidu is probably just to default translation pronouns for genderless, using they, them to call individual might be a bit confusing i guess.
