If I remember correctly from episode 1, monsters are made up of magicules, which is essentially just MP, so anti-magic area should work, I think, and while magicules isn't directly magic it should still weaken them, probably. Someone can probably correct me on this if I'm wrong. Also gotta mention, the sound design is so damn good. And, I'm fairly sure the anime will answer your questions

Anti-magic area doesn't weaken monsters. It just prevents invocation of skills, abilities, or any phenomenon that requires magicules. Clayman instructed Myulan to construct an Anti-Magic Area only because he didn't want the people in Tempest to be able to contact Rimuru while all this shit is happening.

Also yeah, as far as Myulan is aware Clayman is one of the most powerful people in the world and disobedience to him is now not only her death but Youm's, and as Youm said "it's only natural to want to protect what you love" fucking tragic ;-;

There is tho a barrier that weaken monsters, but it's not an Anti-Magic barrier but a Holy barrier (I don't remember how is it called in the anime) that is the one the old man and the other shiro-shouzoku ("white dressed"? I don't know if that's English) conjured at the end

Anti-magic area itself isnt really an issue Myulan knew it too otherwise i believe she would be heavily agains it.. the barrier she casted just prevent of using magic spells including comunicattion.. unfortunately otherworlders dont need it cause they heavily fight with special skills alone which anti magic area dont interfiere.. the issue is the other barrier humans created.. :/ which Myulan didnt know about
